Sampler/Softsynth/Seq...

Can anybody give me any good software out there that can do this. I wanna be able to munipulate my samples, make a melody out of it, edit it, sequence and save it as a .wav file. I have a MPC1K and want and need more flexibility in the near future. Thanks!!! If there's nothing out there that can't do this, this gimme some different software to check out.
